Space Bar Cheat
Once you have built a cannon at the beginning of each level, hold the "space bar" down as the waves of balls pass by the cannon. Keep holding the "space bar" down, and the cannon will consistently track each new wave of balls as they pass by and effectively destroy all of them before they can reach your tower. It may be a good idea to build a few more towers along the route, but this cheat is pretty efficient for most versions of the game, even in the later, harder levels.
Hack and Manipulate the Game
By downloading the program to your desktop, assuming you are using a PC, you can actually manipulate some of the codes within the game to give you more lives or gold (which allows you to build bigger guns to defend your towers). By using a DOS prompt and opening up a file called "flasm" (all easily downloadable online), the code for Tower Defense itself will be available for you to manipulate. Simply search through the code for words like "gold" or "lives." When you find the corresponding number of what you start the game with beside each of those code words, you've found the right strand of code. Simply change those numbers to whatever numbers you would prefer to start with (it usually starts at 80 for gold and 20 for lives), save the program and then restart it from the downloaded file on your computer. Those changes will be in effect in the game.
Unlimited Money
On earlier versions of "Tower Defense" (called "Bloon's Tower Defense"), there is a cheat on the keyboard that will give you unlimited money. Start the game, and once it begins, hold down the keys "Alt" and "F4" at the same time. This will close the window and quit the game. Starting it back up immediately without opening any other applications or restarting the computer should give you unlimited money (gold) to build cannons to defend your towers.
