Unlock Luigi by playing as any character in Adventure Mode. On the first level, time it so your character crosses the finish line when the seconds on the clock are at :02 --- for example, a time of 5 minutes and 2 seconds, 5:02. A clip will appear of Luigi attacking Mario. Once the clip ends, Peach and Luigi will appear to fight. Defeat both of them within 45 seconds without losing a life. Once you've done this, continue through the rest of adventure mode. Once you beat all the levels, Luigi will become unlocked.
Unlock the character Marth by using all 14 default characters in VS Mode. The characters must be controlled by a human, not a computer. Marth can also be locked by playing as all 14 default characters in Classic Mode. Once you've done this, you will be challenged to fight Marth. Beat Marth, and he will become a playable character.
Unlock Mewtwo by playing in VS mode for 20 hours. The VS time is combined for each human player. For example, if two humans play against each other for 10 hours, the game will count the time as 20 hours; if one human plays against a computer for 20 hours, it will count as 20 hours. You can view how much VS time has been played by selecting "Main Menu," then "Data," then "Melee Records," then "Misc. Records." Mewtwo can also be unlocked by playing 700 matches in VS mode. He will then challenge you to a fight. After you beat him, he will be playable.
Unlock Mr. Game &Watch by completing Classic Mode or Adventure Mode on any difficulty setting with every character in the game. This includes all other hidden characters, such as Mewtwo, Luigi, and Dr. Mario. Mr. Game &Watch will then challenge you to a fight. Win the battle, and he will become playable.
