Place your strongest gem in the first tower along the way. You want to cause as much damage to your enemy as soon as possible in order to hold the hordes back.
Create towers at U-shaped bends in the path. This keeps enemies in range of your tower's shots for longer and lets you cause more damage as they wind their way around your tower.
Create high-grade gems and swap them for the lower-grade gems in your towers. One of the new tricks "GemCraft" has come up with is you can place different gems into your towers once you have created them. Create gems that are more powerful as soon as possible and use them to replace the gems you already have in your towers.
Build many towers on boss levels. Bosses are big, tough monsters who will be able to take a lot of punishment. Build many towers and fill them with your most powerful gems to maximize the damage you deal to the boss.
Level up your skills -- especially your mana pool -- as much as possible. The more mana you have, the higher gem grades you can craft and the more towers you can build.
Use your mana early in the game rather than waiting. Your mana will replenish over time but has a maximum cap; so do not let it sit around if you need to use it.
