Unplug the Rock Band guitar wire from the console or computer that it is hooked into. Place the guitar face down. Do this in a dry, well-lit workspace without a lot of dust in the air as you do not want to get any contaminates inside the guitar where they may impede some of the guitar's functions.
Unscrew the screws on the top, or "head," of the guitar first and place them to the side where they will not get lost. The head holds some of the other plastic housings in place so it must be removed before you can access them. Pull the head unit off by hand.
Unscrew the 16 Philips head screws on the back of the Rock Band guitar body with a Phillips head screwdriver. Tape each screw close to the hole that it came from, as the body screws come in separate sizes.
Push a flathead screwdriver gently into the seam along the side of the Rock Band guitar. There may be plastic tabs along the side, depending upon the console version, if so, press the tab back with the flathead. Pull the body housings apart by hand to expose the inner electronic core. The wire comes in at the bottom of the Rock Band guitar and plugs into the electronic circuit board like a normal wire jack. Pull the plug out and lift the wire off the guitar. You can now replace the wire with a new one.
