Put the shoulder strap and guitar over your shoulder, and tighten the strap so you're comfortable and can reach the buttons without stretching too much.
Start a game mode, and select a song with some fast riffs. Choose one you've been struggling with, if you can think of one. Choose a difficulty you're comfortable with.
Place your hands on the fret buttons so your hand covers most of them. Be ready to move your hand down to cover the last button when the notes appear.
Practice double-strumming. On the first strum, push the strum bar down, and on the next note, pull it upward. Repeat this throughout the song, and practice breaking the natural tendency to strum only downward.
Play hammer-ons and pull-offs. When the notes are close together, you can switch the note without having to strum. Press the new fret button quickly, and pull your finger off the old one. For more information on this kind of note, you can find an in-game tutorial which will let you practice the method.
Watch for triple-note chords. You may be familiar with two-note chords, when the game throws two notes at you at the same time. When a chord appears, whether it has two or three notes, press down all of those buttons when you strum.
