Complete missions until you see a scene where Venom and Sandman trap Mary Jane inside a car. This indicates the beginning of the Grand Finale mission. "Spider-Man 3" is not completely linear, so the point this occurs can vary between playthroughs.
Defeat Venom. For most of the fight, Venom is going to be in his rage mode, which is indicated by him glowing red. This makes it difficult to hit him. Hit the metal bars on the second floor. This briefly stuns him, giving you a free shot to do a large amount of damage. Repeat this until he is defeated.
Defeat Sandman. You switch to Harry Osborn for this fight. There is no special trick to this fight. Fly as high as possible to increase the amount of time you have to dodge Sandman's projectiles. Throw bombs at him whenever you get the chance. There are three mini-games that appear during the course of the fight.
Press the button indicated on the screen during the mini-game sequences. Failure to press the button in time results in Harry Osborn losing some life. The boss fight ends once you complete the third mini-game.
Defeat Venom. You switch back to Spider-Man. Both Spider-Man and Venom's health are automatically reduced when the fight starts. This is exactly the same as the previous encounter, except this time you need to hit the pipes above the battle area. This stuns him. A mini-game sequence, like the ones in the Sandman fight, appears after Venom's health is reduced.
Press the buttons when prompted to beat the mini-game. This defeats Venom, completes the Grand Finale mission and unlocks Spider-Man's Venom costume.
